Spatial or organelle proteomics is the systematic study of the proteins and their sub-cellular localization; these compartments can be organelles, i.e. structures defined by lipid bi-layers, macro-molecular assemblies of proteins and nucleic acids or large protein complexes. Highly resolved spatial proteomics has the potential to define the complex cellular and molecular architecture of the human brain in health and disease. (16,17) Most of our approximately 20 000 genes are expressed in the brain in a temporospatial manner, and many demonstrate alternative splicing in different brain regions, hinting at the existence of a very complex, highly compartmentalized Spatial proteomics is the systematic study of protein sub-cellular localisation. We quantify 10,701 proteins as a function of their spatial location and cellular origin. Significant advances in mass spectrometry technologies and proteomic workflows now make proteomics notably more accessible for plant science. Here we describe a systematic mass spectrometry‐based method for spatial proteomics. The approach uses simple fractionation profiling and has two applications: Firstly it can be used to infer subcellular protein localization on a proteome‐wide scale, resulting in a protein map of the cell.
